1. Density Gradient Centrifugation:
In a density gradient preparative ultra-centrifuge, a density gradient must be prepared. This involves layering a lower concentration of sucrose over a higher concentration of sucrose to create a sucrose density gradient. Some ultra-centrifuges are equipped with gradient-forming devices that can generate the gradient automatically. Next, the sample is placed into the centrifuge tube containing the gradient, and the tube is then placed in the rotor of the ultra-centrifuge. Before starting the process, set the temperature and time in the ultra-centrifuge. Close the lid, and the centrifugation process begins. Particles move through the gradient until they reach the point where their density matches that of the surrounding medium. After centrifugation, remove and separate the particles to obtain them as separated units.
2. Differential Centrifugation:
In differential centrifugation, the sample is homogenized in a buffer solution. Then, the sample is placed into a centrifuge tube, which is then run at a specific centrifugal force and temperature for a specific time. At the end of this operation, particles will form at the bottom of the tube, and they are separated from the supernatant. The supernatant is transferred to a new centrifuge tube and centrifuged again at a different speed for a specified time and temperature. Similarly, the supernatant is separated from the formed particles. Continue these steps until all particles are separated from each other.
